DescriptionLAMP1(LysosomalAssociatedMembraneProtein1,alsoknownasCD107a,lysosomalassociatedmembraneglycoprotein1,LGP120andLAMPA)isaproteinprimarilyassociatedwiththelysosomalmembrane.InatypicalcellLAMP1isassociatedwithsphericalvesicleslocatednexttothenucleusandthemicrotubuleorganizingcenter(1).LAMP1isfoundonthecellsurfaceoflymphocytesundergoingdegranulation,aprocessinwhichcytoplasmicvesiclesfusewiththeplasmamembrane,andthisphenomenaresultedindiscoveryofLAMP1asaCDprotein.TheLAMP1proteinhasalargeN-terminalregionwhichisinsidethelysosome,hencetopologicallyexternaltothecell,whichisoftenreferredtoasthelumenaldomain(2).Thelumenaldomainconsistsoftwohomologousglobularsegmentsseparatedbyaprolinerichsequence.Nextthereisasinglemembranespanningdomainandashort11aminoacidC-terminalcytoplasmictail.Thistailregioncontains,attheextremeC-terminus,aso-calledYXXImotifwhichisresponsIBLeforthesortingoftheintactmoleculetotheendosomeandlysozome,whereY=tyrosine,I=isoleucineandX=almostanyaminoacid(3).Thismotifisfoundinseveralotherlysosomalproteins,whereitfunctionsinthesameway.Thereare417aminoacidsinthehumanLAMP1molecule,givinganativemolecularweightof44.8kDa.HowevertheN-terminallumenalsegmentofLAMP1isveryheavilyandvariablyglycosylatedduetothepresenceof18N-linkedglycosylationsites,sothatonSDS-PAGEandonWesternblotstheproteinrunsasadiffusebandat90-120kDa.AntibodiestoLAMP1arethereforeexcellentMarkersoflysosomesinmammaliancells,thoughsomeLAMP1mayalsobeseenonlateendosomesandontheplasmamembrane.
